Locin 500 mg tablet contains Levofloxacin, a broad-spectrum fluoroquinolone antibiotic used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Usually 500 mg once daily for 7-14 days depending on the infection type and severity.
Elderly
Dosage adjustment may be necessary in elderly patients with impaired renal function.

Dosage adjustment required based on creatinine clearance (e.g., lower dose or extended interval for CrCl < 50 mL/min).
How to Take
May be taken with or without food. Swallow the tablet whole with water. Do not crush or chew.
Mechanism of Action
Levofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, enzymes essential for bacterial DNA replication, transcription, repair, and recombination, leading to bacterial cell death.

Excretion
Primarily excreted unchanged in the urine, mainly via glomerular filtration and tubular secretion.

Minimally metabolized; primarily excreted unchanged.
Side Effects
Contraindications
- Hypersensitivity to levofloxacin or other fluoroquinolones
- History of tendinitis or tendon rupture associated with fluoroquinolone use
- Epilepsy
- Children and growing adolescents (due to risk of cartilage damage)
- Pregnancy and lactation (unless benefits outweigh risks)
Drug Interactions
NSAIDs
May increase risk of CNS stimulation and convulsions.
Warfarin
May enhance anticoagulant effect, increasing bleeding risk. Monitor INR.
Corticosteroids
Increased risk of tendinopathy and tendon rupture.
Antiarrhythmics (e.g., amiodarone, quinidine)
Increased risk of QT prolongation.
Antacids (containing magnesium/aluminum), sucralfate, iron supplements, zinc supplements
Reduce absorption of levofloxacin; take levofloxacin at least 2 hours before or 2 hours after these agents.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

Symptoms may include CNS effects (confusion, tremors, seizures), QT prolongation. Treatment involves symptomatic and supportive measures. Gastric lavage and hydration may be helpful. Hemodialysis is not effective.
