Description
Overview of the medicine
Loratadine is a non-drowsy antihistamine used to relieve allergy symptoms such as sneezing, runny nose, itchy or watery eyes, and itching of the nose or throat.
Uses & Indications
Dosage
Adults
10 mg once daily
Elderly
No dosage adjustment generally required, but caution in severe hepatic impairment.
Renal_impairment
For adults and children 6 years of age and over with liver or kidney failure, the initial dose should be 10 mg every other day.
How to Take
Administer orally, with or without food.
Mechanism of Action
Loratadine is a tricyclic antihistamine with selective peripheral H1-receptor antagonist activity. It prevents histamine from binding to H1 receptors, thereby alleviating allergy symptoms without significant sedation due to its poor penetration of the blood-brain barrier.

1-3 hours
Excretion
Approximately 40% excreted in urine and 42% in feces over 10 days, primarily as conjugated metabolites.
Half life
Loratadine: 8.4 hours; active metabolite (desloratadine): 28 hours.
Absorption
Rapidly absorbed after oral administration; peak plasma concentrations reached in 1-2 hours.
Metabolism
Extensively metabolized in the liver by cytochrome P450 enzymes (CYP3A4 and CYP2D6) to descarboethoxyloratadine (desloratadine), an active metabolite.

Cimetidine
Increased plasma concentrations of loratadine and desloratadine.
Erythromycin
Increased plasma concentrations of loratadine and desloratadine.
Ketoconazole
Increased plasma concentrations of loratadine and desloratadine.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include somnolence, tachycardia, and headache. Gastric lavage and symptomatic/supportive treatment recommended.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed. Excreted into breast milk, not recommended during lactation.
