Overview of the medicine
Lorfast 10 mg Tablet contains Loratadine, a second-generation antihistamine. It is used to relieve symptoms associated with allergic rhinitis (like sneezing, runny or itchy nose, itchy watery eyes) and chronic idiopathic urticaria (hives). It is known for its non-sedating properties.
Uses & Indications
Dosage
Adults
10 mg once daily.
Elderly
No specific dose adjustment is generally required for elderly patients.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min) or hepatic impairment, a starting dose of 10 mg every other day is recommended.
How to Take
Take Lorfast 10 mg Tablet orally, with or without food, as directed by your doctor or pharmacist. Swallow the tablet whole with water.
Mechanism of Action
Loratadine is a selective peripheral H1-receptor antagonist. It works by blocking the action of histamine, a natural substance in the body that causes allergic symptoms. By blocking H1 receptors, it prevents histamine from binding and exerting its effects, thereby reducing allergic reactions.

Onset
Onset of action typically occurs within 1 to 3 hours.
Excretion
Approximately 80% of the dose is excreted equally in the urine and feces as metabolic products within 10 days.
Half life
Loratadine: 8 to 15 hours; Descarboethoxyloratadine (active metabolite): 17 to 24 hours.
Absorption
Rapidly and well absorbed from the gastrointestinal tract after oral administration.
Metabolism
Extensively metabolized in the liver, primarily by CYP2D6 and CYP3A4, to its active metabolite, descarboethoxyloratadine.
Side Effects
Contraindications
- Hypersensitivity to Loratadine, desloratadine, or any component of the formulation.
- Severe hepatic impairment (caution required, dose adjustment may be necessary).
Drug Interactions
Cimetidine
May increase plasma concentrations of loratadine and descarboethoxyloratadine.
Erythromycin
May increase plasma concentrations of loratadine and descarboethoxyloratadine.
Ketoconazole
May increase plasma concentrations of loratadine and descarboethoxyloratadine.

Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

In adults, somnolence, tachycardia, and headache have been reported. In the event of an overdose, symptomatic and supportive treatment should be initiated immediately. Gastric lavage may be considered if appropriate. Loratadine is not effectively removed by hemodialysis.

Pregnancy Category B. Loratadine is excreted in breast milk; therefore, caution should be exercised when administered to a nursing woman. Consult your doctor before use during pregnancy or lactation.
