Description
Overview of the medicine
Loratadine is a non-sedating antihistamine used to relieve symptoms of seasonal allergic rhinitis and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
For adults and children 6 years and older: 10 ml (10 mg) once daily.
Elderly
No dosage adjustment needed for elderly patients with normal renal and hepatic function.
Renal_impairment
For severe renal impairment (GFR < 30 mL/min): 10 ml (10 mg) every other day.
How to Take
Shake well before use. Take orally with or without food. Use the provided measuring device to ensure accurate dosing.
Mechanism of Action
Loratadine selectively antagonizes peripheral H1-receptors, thereby inhibiting the effects of histamine.
Pharmacokinetics
Onset
Within 1-3 hours.
Excretion
Approximately 40% in urine and 42% in feces over 10 days, mainly as conjugated metabolites.
Half life
Loratadine: 8.4 hours (range 3-20 hours); Descarboethoxyloratadine (active metabolite): 12-24 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations reached in 1-1.5 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP2D6 and CYP3A4, to descarboethoxyloratadine.
Side Effects
Contraindications
- •Hypersensitivity to Loratadine or any component of the formulation.
Drug Interactions
Cimetidine
Concomitant use may increase plasma concentrations of loratadine and its active metabolite. Monitor for increased side effects.
Erythromycin
Concomitant use may increase plasma concentrations of loratadine and its active metabolite. Monitor for increased side effects.
Ketoconazole
Concomitant use may increase plasma concentrations of loratadine and its active metabolite. Monitor for increased side effects.
Other CYP3A4 or CYP2D6 inhibitors
Potential for increased loratadine levels; caution advised.
Storage
Store below 30°C in a dry place, protected from light and moisture. Do not freeze.
Overdose
Symptoms of overdose include somnolence, tachycardia, and headache. In case of overdose, initiate general symptomatic and supportive measures. Gastric lavage may be considered if appropriate. Loratadine is not effectively removed by hemodialysis.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown a risk, but human studies are inadequate. Use only if clearly needed. Loratadine and its active metabolite are excreted in breast milk; caution is advised when administered to a nursing woman.
