Description
Overview of the medicine
Magasol is a magnesium supplement used to treat and prevent magnesium deficiency. Magnesium is an essential mineral required for various bodily functions including nerve and muscle function, blood glucose control, blood pressure regulation, and bone health.
Uses & Indications
Dosage
Adults
Typically 1-2 tablets (500-1000 mg) daily, preferably with food, or as directed by a physician.
Elderly
No specific dose adjustment for elderly patients, but caution is advised in those with impaired renal function.
Renal_impairment
Dose reduction may be necessary in patients with moderate to severe renal impairment due to the risk of hypermagnesemia. Consult a physician.
How to Take
Take orally with a glass of water, preferably with a meal to minimize gastrointestinal upset. Do not crush or chew the tablet unless it is an effervescent or chewable form.
Mechanism of Action
Magnesium acts as a cofactor in over 300 enzymatic reactions, including those involved in ATP synthesis, muscle contraction, and nerve signal transmission. It also plays a role in nucleic acid and protein synthesis, and maintaining electrolyte balance.
Pharmacokinetics
Onset
Clinical effects for deficiency symptoms may take hours to days or weeks to become noticeable, depending on the severity of the deficiency.
Excretion
Primarily excreted by the kidneys. The kidneys regulate magnesium balance by adjusting excretion based on dietary intake and body needs.
Half life
The half-life of magnesium in plasma is short, but total body magnesium turnover is much slower due to storage in bones and soft tissues.
Absorption
Magnesium absorption primarily occurs in the small intestine (jejunum and ileum) via both passive and active transport mechanisms. Bioavailability varies depending on the specific salt and individual factors.
Metabolism
Magnesium is not metabolized by the body. It functions in its ionic form.
Side Effects
Contraindications
- •Hypersensitivity to magnesium L-aspartate or any component of the formulation
- •Severe renal impairment (creatinine clearance < 30 mL/min)
- •Heart block (e.g., AV block)
- •Myocardial damage
- •Hypermagnesemia
Drug Interactions
Bisphosphonates
Reduced absorption of bisphosphonates. Administer at least 30 minutes apart.
Calcium channel blockers
Potential for additive hypotensive effects.
Diuretics (loop and thiazide)
May alter magnesium excretion, potentially leading to hypomagnesemia or hypermagnesemia depending on the diuretic type.
Tetracyclines & Quinolone antibiotics
Magnesium can chelate with these antibiotics, reducing their absorption. Administer at least 2-4 hours apart.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of magnesium overdose (hypermagnesemia) include diarrhea, nausea, vomiting, muscle weakness, lethargy, decreased reflexes, hypotension, and in severe cases, respiratory depression, cardiac arrest, and coma. Management involves intravenous calcium gluconate to counteract cardiotoxicity, IV fluids to promote renal excretion, and in severe cases, hemodialysis.
Pregnancy & Lactation
Magnesium is essential during pregnancy and lactation. Magasol is generally considered safe when taken at recommended doses, but always consult a physician before use. Excessively high doses should be avoided.
