Description
Overview of the medicine
Meroclav 500 mg Tablet is an antibiotic medication combining Amoxicillin, a penicillin-type antibiotic, and Clavulanic Acid, a beta-lactamase inhibitor. It is used to treat a wide range of bacterial infections susceptible to this combination.
Uses & Indications
Dosage
Adults
One 500 mg/125 mg tablet every 8 hours, or as directed by physician. For more severe infections, dosage may be increased.
Elderly
Same as adults, but caution with renal impairment; dosage adjustment may be needed.
Renal_impairment
Dosage adjustment required based on creatinine clearance. For CrCl 10-30 mL/min, 500 mg/125 mg every 12 hours; for CrCl <10 mL/min, 500 mg/125 mg every 24 hours.
How to Take
Taken orally at the start of a meal to minimize potential gastrointestinal intolerance. Swallow whole with water.
Mechanism of Action
Amoxicillin acts by inhibiting bacterial cell wall synthesis, leading to bacterial lysis and death. Clavulanic acid inhibits beta-lactamase enzymes produced by some bacteria, which would otherwise inactivate Amoxicillin, thus extending Amoxicillin's antibacterial spectrum.
Pharmacokinetics
Onset
Within 1-2 hours (antibacterial activity)
Excretion
Primarily excreted unchanged by the kidneys (glomerular filtration and tubular secretion).
Half life
Approximately 1-1.5 hours for both components.
Absorption
Both Amoxicillin and Clavulanic Acid are well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Amoxicillin is minimally metabolized; Clavulanic Acid is extensively metabolized.
Side Effects
Contraindications
- History of hypersensitivity to Amoxicillin, Clavulanic Acid, or any penicillin.
- History of cholestatic jaundice/hepatic dysfunction associated with Amoxicillin/Clavulanic Acid.
Drug Interactions
Warfarin
May prolong prothrombin time; monitor INR closely.
Probenecid
Increases and prolongs Amoxicillin blood levels.
Allopurinol
Increased incidence of rash.
Methotrexate
May increase methotrexate toxicity by inhibiting its renal excretion.
Oral Contraceptives
May reduce efficacy of oral contraceptives; consider alternative contraception.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms may include gastrointestinal upset and fluid/electrolyte imbalance. Treatment is symptomatic and supportive. Hemodialysis may be used to remove the drug from circulation.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe, but use with caution and only if clearly needed. Excreted in breast milk, monitor infants for diarrhea, candidiasis, and skin rash.
