Overview of the medicine
Miganil (Pizotifen) is an antihistamine and serotonin antagonist used for the prophylaxis of migraine and other vascular headaches. It may also have appetite stimulant properties.
Uses & Indications
Dosage
Adults
Normally, 0.5-1.5 mg daily, gradually increased to 1.5-3 mg daily in divided doses or as a single dose at night. A single 5 mg tablet (if prescribed) would be considered a high daily dose and should be taken with extreme caution, likely once daily at night, under strict medical supervision. Maximum recommended daily dose is typically 4.5 mg.
Elderly
Start with lower doses (e.g., 0.5 mg daily) and titrate carefully due to increased sensitivity to side effects.
Renal_impairment
Use with caution. Dosage reduction might be necessary due to altered excretion.
How to Take
Oral administration. Can be taken with or without food. To minimize sedation, especially with higher doses like 5mg, it is often recommended to take the dose at night. Start with a low dose and gradually increase.
Mechanism of Action
Pizotifen acts as a serotonin (5-HT1 and 5-HT2) antagonist and has antihistaminic (H1) and weak anticholinergic properties. It is believed to work in migraine prophylaxis by reducing serotonin-induced vasoconstriction and by stabilizing mast cells. It also increases appetite.
