Description
Overview of the medicine
Mosapride is a gastroprokinetic agent used to treat symptoms of gastroesophageal reflux disease and functional dyspepsia, such as heartburn, nausea, vomiting, and bloating.
Uses & Indications
Dosage
Adults
Usually 5 mg three times daily before or after meals, or 10 mg once or twice daily, as directed by a physician. Maximum recommended dose is typically 15 mg/day.
Elderly
Lower doses may be considered, and patients should be monitored for adverse effects. Consult a physician.
Renal_impairment
Use with caution. Dose adjustment may be necessary in severe renal impairment. Consult a physician.
How to Take
Take orally with a glass of water, with or without food, as prescribed by your physician. It is often recommended to take before meals for optimal effect.
Mechanism of Action
Mosapride selectively acts as a 5-HT4 receptor agonist, which increases acetylcholine release from enteric neurons. This enhanced acetylcholine leads to increased gastrointestinal motility and accelerated gastric emptying.
Pharmacokinetics
Onset
Within 30 minutes to 1 hour.
Excretion
Excreted mainly via urine and feces, with metabolites being the primary form.
Half life
Approximately 1-2 hours for the parent drug and longer for active metabolites.
Absorption
Rapidly absorbed after oral administration, peak plasma concentration reached in approximately 1-2 hours.
Metabolism
Primarily metabolized in the liver, mainly by CYP3A4, to its active metabolite M-1.
Side Effects
Contraindications
- •Hypersensitivity to mosapride or any component of the formulation.
- •Patients with gastrointestinal hemorrhage, mechanical obstruction, or perforation.
Drug Interactions
Erythromycin
May increase mosapride plasma concentrations due to CYP3A4 inhibition, potentially leading to increased adverse effects.
Anticholinergic drugs
May decrease the prokinetic effect of mosapride.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include increased gastrointestinal side effects. Management is symptomatic and supportive. There is no specific antidote.
Pregnancy & Lactation
Use with caution during pregnancy and lactation only if clearly needed and potential benefits outweigh the risks. Animal studies have shown no significant teratogenic effects. Consult a doctor.
