Description
Overview of the medicine
Myobac 10 mg Tablet contains Baclofen, a centrally acting skeletal muscle relaxant used to treat muscle spasms, rigidity, and pain caused by various neurological conditions.
Uses & Indications
Dosage
Adults
Initial dose is usually 5 mg three times daily, gradually increased by 5 mg every 3 days to a maximum of 20 mg four times daily (80 mg/day).
Elderly
Lower initial doses (e.g., 5 mg once or twice daily) are recommended, with careful titration due to increased sensitivity and risk of side effects.
Renal_impairment
Dosage reduction is required. For severe renal impairment, consider 5 mg daily or every other day, monitoring for adverse effects.
How to Take
Take orally with food or milk to minimize gastrointestinal upset. Do not discontinue abruptly.
Mechanism of Action
Baclofen acts as a GABA-B receptor agonist, leading to hyperpolarization of neurons and reduced release of excitatory neurotransmitters. This inhibits both monosynaptic and polysynaptic reflexes at the spinal cord level, thereby reducing muscle spasms and spasticity.
Pharmacokinetics
Onset
Onset of action typically occurs within 3-4 hours.
Excretion
Primarily excreted unchanged by the kidneys (approx. 70-80% within 72 hours).
Half life
2.5 to 4 hours (elimination half-life).
Absorption
Rapidly and extensively absorbed from the gastrointestinal tract. Bioavailability is about 70-85%.
Metabolism
Minimal hepatic metabolism (approx. 5-15% of dose).
Side Effects
Contraindications
- •Hypersensitivity to Baclofen or any component of the formulation.
- •Epilepsy (uncontrolled).
Drug Interactions
Antihypertensives
Additive hypotensive effects may occur.
Tricyclic Antidepressants
May potentiate baclofen's muscle relaxant effect.
Alcohol and other CNS depressants
Increased central nervous system depression (drowsiness, sedation).
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms include severe drowsiness, dizziness, muscle weakness, respiratory depression, seizures, and coma. Treatment is supportive, focusing on maintaining vital functions.
Pregnancy & Lactation
Use in pregnancy only if the potential benefit outweighs the potential risk to the fetus. Excreted in breast milk; use with caution during lactation, consult a doctor.
