For oral candidiasis, the tablet should be slowly dissolved in the mouth or held in the mouth for as long as possible before swallowing, to allow prolonged contact with the affected areas. For intestinal candidiasis, the tablet should be swallowed whole.

Nystatin binds to ergosterol, a primary component of the fungal cell membrane, leading to the formation of pores and increased permeability. This results in leakage of intracellular components like potassium and other essential nutrients, ultimately causing fungal cell death.

Nystatin is generally well-tolerated even in high doses due to its poor systemic absorption. Oral doses of 5 million units daily for several weeks have been tolerated without serious adverse effects. Gastric upset, nausea, and diarrhea may occur.

Pregnancy Category B. Nystatin is generally considered safe during pregnancy as it is not significantly absorbed from the GI tract. Consult a doctor before use. It is unknown whether Nystatin is excreted in human milk, but due to poor oral absorption, it is generally considered compatible with breastfeeding.
