Description
Overview of the medicine
Naspro 250 mg Tablet contains Naproxen, a non-steroidal anti-inflammatory drug (NSAID) used for the relief of pain, inflammation, and fever.
Uses & Indications
Dosage
Adults
Typically 250-500 mg twice daily. Maximum 1250 mg/day for short periods.
Elderly
Lower doses may be required; closely monitor for adverse effects due to increased risk of side effects.
Renal_impairment
Use with caution and at reduced doses. Avoid in severe renal impairment (creatinine clearance < 30 mL/min).
How to Take
Take Naspro 250 mg Tablet orally, preferably with food or milk to reduce gastrointestinal upset. Swallow the tablet whole with a glass of water, do not crush or chew.
Mechanism of Action
Naproxen inhibits cyclooxygenase (COX-1 and COX-2) enzymes, thereby blocking prostaglandin synthesis, which are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Approximately 1 hour for pain relief.
Excretion
Primarily renal (about 95% as naproxen and its metabolites), with a small amount excreted in feces.
Half life
12-17 hours.
Absorption
Rapid and complete absorption from the gastrointestinal (GI) tract. Peak plasma levels usually occur within 2-4 hours after oral administration.
Metabolism
Primarily hepatic via the cytochrome P450 (CYP450) system to inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to naproxen, aspirin, or other NSAIDs.
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs.
- Peri-operative pain in the setting of coronary artery bypass graft (CABG) surgery.
- Active gastrointestinal bleeding or peptic ulcer disease.
- Severe heart failure, renal or hepatic impairment.
Drug Interactions
Lithium
Increased plasma lithium levels, leading to toxicity.
SSRIs/SNRIs
Increased risk of gastrointestinal bleeding.
Methotrexate
Increased methotrexate toxicity.
ACE inhibitors/ARBs
Increased risk of renal impairment and reduced antihypertensive effect.
Diuretics (e.g., Furosemide)
Reduced diuretic and antihypertensive effects.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from direct sunlight, heat, and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, heartburn, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Management involves symptomatic and supportive care. Gastric lavage or activated charcoal may be considered if recent ingestion.
Pregnancy & Lactation
Pregnancy Category C (Category D in the third trimester or near delivery). Avoid in late pregnancy due to the risk of premature closure of the fetal ductus arteriosus. Use with caution during lactation; small amounts are excreted in breast milk.
