Neofix 100 mg Suspension contains Cefixime, a third-generation cephalosporin antibiotic used to treat a wide range of bacterial infections. It is effective against various Gram-positive and Gram-negative bacteria.
Uses & Indications
Dosage
Adults
This 100 mg/5 ml suspension is primarily formulated for pediatric use. For adult Cefixime, typical dose is 200-400 mg daily (as single or two divided doses), usually in tablet/capsule form. For children (6 months to 12 years): 8 mg/kg/day, given as a single daily dose or in two divided doses. For Neofix 100 mg/5ml Suspension: 6 months-1 year: 3.75 ml once daily; 1-4 years: 5 ml once daily; 5-8 years: 10 ml once daily; 9-12 years: 15 ml once daily. Or as directed by physician.
Elderly
No specific dosage adjustment is required for elderly patients based on age alone, but renal function should be assessed and dose adjusted accordingly. Consult physician.
Renal_impairment
For creatinine clearance 21-60 mL/min, 75% of the standard daily dose. For creatinine clearance < 20 mL/min, 50% of the standard daily dose. Consult physician.
How to Take
Administer orally. Shake the bottle well before each use. Can be taken with or without food.
Mechanism of Action
Cefixime is a bactericidal antibiotic that acts by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located inside the bacterial cell wall, which prevents the cross-linking of peptidoglycan chains, leading to cell lysis and bacterial death.
Pharmacokinetics
Onset
Peak plasma concentrations usually reached within 2-6 hours.
Excretion
Primarily excreted by the kidneys (50-60% as unchanged drug) and also via biliary excretion.
Half life
Approximately 3-4 hours.
Absorption
Approximately 40-50% orally absorbed. Food may delay absorption but does not significantly affect the extent.
Metabolism
Minimal hepatic metabolism.
Side Effects
Contraindications
- Hypersensitivity to Cefixime, other cephalosporins, or penicillins.
- Patients with a history of severe allergic reactions to any beta-lactam antibiotic.
Drug Interactions
Warfarin
May increase prothrombin time (PT) and INR, increasing risk of bleeding. Close monitoring required.
Carbamazepine
Increased carbamazepine levels reported with concomitant use. Monitor carbamazepine levels.
Oral Contraceptives
May decrease the efficacy of oral contraceptives. Advise patients to use an alternative or additional method of contraception.
Storage
Store dry powder below 30°C, protected from light and moisture. Keep reconstituted suspension in a refrigerator (2-8°C) and discard after 7 days.
Overdose
In case of overdose, gastric lavage may be indicated. There is no specific antidote. Treatment should be symptomatic and supportive. Hemodialysis or peritoneal dialysis does not remove significant quantities of cefixime.
