Neurolin-CR 165 mg Tablet is a controlled-release formulation of Pregabalin, primarily used for the management of neuropathic pain, fibromyalgia, and as an adjunctive therapy for partial onset seizures.
Uses & Indications
Dosage
Adults
For neuropathic pain, typically initiated with 165 mg once daily. Dosage may be increased based on response and tolerability, up to a maximum of 330 mg once daily. For fibromyalgia, start with 165 mg once daily, may increase to 330 mg once daily.
Elderly
Dose adjustments may be necessary due to age-related decline in renal function. Start with a lower dose and titrate cautiously.
Renal_impairment
Significant dose adjustment is required based on creatinine clearance. For CrCl 30-60 mL/min, 82.5 mg once daily. For CrCl <30 mL/min, 50 mg every other day.
How to Take
Take orally with or without food. Swallow the tablet whole; do not crush, chew, or split it.
Mechanism of Action
Pregabalin binds with high affinity to the alpha2-delta (α2-δ) subunit of voltage-gated calcium channels in the central nervous system. This binding modulates the release of several excitatory neurotransmitters, thereby exerting its analgesic, anxiolytic, and anticonvulsant effects.

Analgesic effects for neuropathic pain may be observed within one week of initiation.
Excretion
Primarily excreted unchanged in the urine (approximately 98% of the dose).
Half life
Approximately 6.3 hours. The controlled-release formulation extends the duration of action.
Absorption
Rapidly absorbed after oral administration, with controlled-release formulations designed for sustained absorption. Bioavailability is ≥90% and is not affected by food.
Metabolism
Negligible metabolism in humans; less than 0.1% of the dose is metabolized.
Side Effects
Contraindications
- Hypersensitivity to pregabalin or any component of the formulation.
Drug Interactions
CNS Depressants (e.g., opioids, benzodiazepines, alcohol)
May enhance the central nervous system (CNS) depressant effects, including sedation and somnolence.
Thiazolidinedione Antidiabetics (e.g., rosiglitazone, pioglitazone)
May increase the risk of edema and weight gain, especially in patients with pre-existing cardiac conditions.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and in rare cases, coma. Treatment is supportive and symptomatic; hemodialysis may be considered in severe cases as pregabalin is hemodialyzable.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Pregabalin is excreted in human milk; caution is advised when administered to a nursing woman.
