Description
Overview of the medicine
Nizox 100 mg Suspension is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, reduce fever, and manage inflammation.
Uses & Indications
Dosage
Adults
Not typically indicated for adult use in 100mg suspension form. Adult dosage for tablets is usually 100 mg twice daily.
Elderly
Use with caution. A reduced dosage may be necessary, especially in frail patients or those with impaired renal or hepatic function.
Children
5 mg/kg body weight per day in 2 or 3 divided doses. Max 100 mg per day.
Renal_impairment
In patients with mild to moderate renal impairment, dose adjustment may be necessary. Contraindicated in severe renal impairment.
How to Take
Administer orally, preferably after food, to minimize gastrointestinal discomfort. Use the provided measuring device for accurate dosing.
Mechanism of Action
Nimesulide works by inhibiting the synthesis of prostaglandins, primarily by selectively inhibiting the cyclooxygenase (COX)-2 enzyme, which is responsible for mediating inflammation, pain, and fever.
Pharmacokinetics
Onset
Analgesic effect usually within 30-60 minutes.
Excretion
Approximately 65% of the dose is excreted in the urine and 35% in the feces.
Half life
Elimination half-life ranges from 2 to 5 hours.
Absorption
Rapidly and well absorbed after oral administration, with peak plasma concentrations reached within 1.5-3 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C9, into its main active metabolite, 4'-hydroxynimesulide.
Side Effects
Contraindications
- Hypersensitivity to nimesulide or any component of the formulation
- History of gastrointestinal bleeding or peptic ulcer
- Severe hepatic impairment
- Severe renal impairment
- Children under 12 years of age (official guidelines, though used off-label in some regions)
- Third trimester of pregnancy and lactation
Drug Interactions
Lithium
Nimesulide may increase plasma lithium levels.
Cyclosporine
Increased risk of nephrotoxicity.
Methotrexate
May increase methotrexate toxicity.
Diuretics and ACE inhibitors
May reduce the antihypertensive effect and increase the risk of renal impairment.
Warfarin and other anticoagulants
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms may include lethargy, drowsiness, epigastric pain, nausea, vomiting, and gastrointestinal bleeding. Management is supportive and symptomatic; includes gastric lavage, activated charcoal, and monitoring of vital signs and liver function.
Pregnancy & Lactation
Contraindicated in the third trimester of pregnancy due to potential fetal harm (e.g., premature closure of the ductus arteriosus). Avoid use during the first two trimesters and lactation unless absolutely necessary and potential benefits outweigh risks.

- Renal function tests
- Complete Blood Count (CBC) if used for prolonged periods.
Doctor Notes
- Exercise caution when prescribing to patients with a history of gastrointestinal disorders, renal impairment, or hepatic dysfunction.
- Monitor liver function tests regularly, especially during prolonged treatment or in high-risk patients.
