Dosage

500 mg orally every 12 hours for 3 days, taken with food.

Mechanism of Action
Nitazoxanide interferes with the pyruvate ferredoxin oxidoreductase (PFOR) enzyme-dependent electron transfer reaction, which is essential for anaerobic energy metabolism in certain parasites.

Excretion
Primarily excreted in feces (approximately 60%) as tizoxanide and tizoxanide glucuronide, and to a lesser extent in urine (less than 1%) as tizoxanide.
Half life
Plasma half-life of tizoxanide is approximately 1.5 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract and rapidly hydrolyzed to its active metabolite, tizoxanide, and tizoxanide glucuronide.
Metabolism
Extensively metabolized by hydrolysis to tizoxanide and tizoxanide glucuronide.
Side Effects
Contraindications
- Hypersensitivity to nitazoxanide or any component of the formulation.
- Patients with severe hepatic impairment.
Drug Interactions
Warfarin
Nitazoxanide may displace warfarin from plasma protein binding sites, leading to an increased prothrombin time (INR). Monitor INR closely.
Highly protein-bound drugs
Caution should be exercised when nitazoxanide is co-administered with other highly protein-bound drugs with narrow therapeutic indices, as competition for binding sites may occur.

Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
