Overview of the medicine
Ofpain 10 mg Tablet is a non-steroidal anti-inflammatory drug (NSAID) used for the short-term management of moderate to severe acute pain. It works by blocking certain natural substances in the body that cause inflammation and pain.
Uses & Indications
Dosage
Adults
Initially 10 mg orally, then 10-20 mg every 4-6 hours as needed. Maximum 40 mg/day for not more than 5 days.
Elderly
Lower doses (e.g., 10 mg every 6-8 hours) may be required due to decreased renal function and increased risk of adverse effects.
Renal_impairment
Dose adjustment necessary; contra-indicated in severe renal impairment.
How to Take
Take with food or milk to minimize gastrointestinal upset. Do not chew or crush the tablet. Swallow whole with water.
Mechanism of Action
Ketorolac inhibits prostaglandin synthesis by reversibly blocking the cyclooxygenase (COX-1 and COX-2) enzymes. This results in reduced formation of prostaglandin precursors, which are involved in inflammation, pain, and fever.

- Active peptic ulcer, GI bleeding

- Advanced heart failure
- Prior to major surgery

- History of asthma, urticaria, or allergic-type reactions after aspirin or other NSAIDs
Drug Interactions
Lithium
Increased lithium plasma levels.
Methotrexate
Increased methotrexate toxicity.
Warfarin, Heparin
Increased risk of bleeding.
Diuretics (Furosemide)
Reduced diuretic and antihypertensive effects.
ACE inhibitors, Angiotensin II receptor blockers
Increased risk of renal impairment.

Overdose
Symptoms may include drowsiness, nausea, vomiting, epigastric pain, GI bleeding, and rarely, acute renal failure, hypertension, and respiratory depression. Treatment is symptomatic and supportive; no specific antidote.
Pregnancy & Lactation
Not recommended during the third trimester of pregnancy due to risk of premature closure of fetal ductus arteriosus. Use during lactation is generally discouraged as NSAIDs are excreted in breast milk.
