Description
Overview of the medicine
Oncodex 8 mg Tablet is an oral targeted therapy used in the treatment of certain types of cancer, particularly those driven by specific genetic mutations. It works by inhibiting key enzymes involved in cancer cell growth and survival.
Uses & Indications
Dosage
Adults
The usual recommended dose is 8 mg orally once daily. Dosage may be adjusted based on individual response and tolerability.
Elderly
No specific dose adjustment is required for elderly patients, but careful monitoring for adverse effects is recommended.
Renal_impairment
No dose adjustment needed for mild to moderate renal impairment. Use with caution in severe renal impairment (CrCl <30 mL/min) with close monitoring.
How to Take
Take Oncodex 8 mg Tablet orally once daily, with or without food, at approximately the same time each day. Swallow the tablet whole; do not crush, chew, or split it.
Mechanism of Action
Targetinib, the active ingredient in Oncodex, is a selective tyrosine kinase inhibitor. It targets and inhibits specific receptor tyrosine kinases (e.g., EGFR, ALK, ROS1 depending on the specific product profile it mimics) that are overexpressed or mutated in certain cancer cells, thereby blocking downstream signaling pathways essential for tumor growth, proliferation, and survival.
Pharmacokinetics
Onset
Clinical effects may be observed within weeks, though anti-tumor activity can begin earlier at a cellular level.
Excretion
Excreted predominantly in feces (approx. 70-80%) with a smaller portion in urine (approx. 10-20%).
Half life
Elimination half-life typically ranges from 18-24 hours, allowing for once-daily dosing.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations reached within 2-4 hours. Bioavailability is approximately 60-70%.
Metabolism
Primarily metabolized in the liver by cytochrome P450 enzymes, mainly CYP3A4.
Side Effects
Contraindications
- •Known hypersensitivity to Targetinib or any excipients in the tablet.
- •Severe hepatic impairment.
Drug Interactions
Strong CYP3A4 inducers (e.g., Rifampicin, Carbamazepine)
May decrease Targetinib exposure; avoid co-administration or increase Oncodex dose with caution.
Strong CYP3A4 inhibitors (e.g., Ketoconazole, Ritonavir)
May increase Targetinib exposure; reduce Oncodex dose if co-administration is necessary.
Proton pump inhibitors (PPIs) and H2-receptor antagonists
May decrease Targetinib absorption; consider separating administration times or using alternative acid-reducing agents.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
In case of an overdose, there is no specific antidote. Treatment should consist of general supportive measures. Closely monitor patients for signs and symptoms of toxicity and manage symptomatically. Consider gastrointestinal decontamination if appropriate.
Pregnancy & Lactation
Oncodex is not recommended during pregnancy as it may cause fetal harm. Effective contraception should be used by women of reproductive potential during treatment and for at least 6 months after the last dose. It is unknown whether Targetinib is excreted in human milk; due to the potential for serious adverse reactions in breastfed infants, women should not breastfeed during treatment and for at least 2 weeks after the last dose.
