Onride 8 mg Tablet contains Ondansetron, an antiemetic drug used to prevent nausea and vomiting caused by chemotherapy, radiation therapy, and surgery. It works by blocking the action of serotonin, a natural substance in the body that can cause nausea and vomiting.
Uses & Indications
Dosage
Adults
Typically 8 mg orally twice daily, or as directed by physician for prevention of chemotherapy-induced nausea and vomiting or postoperative nausea and vomiting.
Elderly
No specific dose adjustment for elderly, but careful monitoring advised.
Renal_impairment
No dose adjustment is required for patients with renal impairment.
How to Take
Onride 8 mg Tablet should be taken orally with or without food. For chemotherapy-induced nausea and vomiting, take the first dose 30 minutes before the start of chemotherapy. For postoperative nausea and vomiting, take 1 hour before anesthesia or as directed by the physician.
Mechanism of Action
Ondansetron is a selective 5-HT3 receptor antagonist. Serotonin 5-HT3 receptors are located peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone (CTZ). By blocking these receptors, Ondansetron prevents the initiation of the vomiting reflex stimulated by chemotherapy or radiation, or post-surgical events.
Pharmacokinetics
Onset
Within 30 minutes to 2 hours after oral administration.
Excretion
About 5% of the dose is excreted unchanged in urine, with the remainder excreted as metabolites via urine and feces.
Half life
Approximately 3-5 hours in adults.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract, with peak plasma concentrations occurring approximately 1.5 hours after oral administration.
Metabolism
Extensive hepatic metabolism via hydroxylation, followed by glucuronide or sulfate conjugation.
Side Effects
Contraindications
- Hypersensitivity to Ondansetron or any component of the formulation
- Concomitant use with apomorphine (due to risk of profound hypotension and loss of consciousness)
Drug Interactions
Tramadol
May reduce the analgesic effect of tramadol.
Apomorphine
Concomitant use is contraindicated due to reports of profound hypotension and loss of consciousness.
Serotonergic drugs (SSRIs, SNRIs)
Increased risk of serotonin syndrome.
Phenytoin, Carbamazepine, Rifampicin
May decrease ondansetron levels.
Storage
Store at room temperature (below 30°C), away from light and moisture. Keep out of reach of children.

Symptoms of overdose include transient visual disturbances, severe constipation, hypotension, and vasovagal episodes with transient AV block. Treatment is symptomatic and supportive. There is no specific antidote.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus, but there are no adequate and well-controlled studies in pregnant women. Use during pregnancy only if clearly needed. Excretion into breast milk is unknown; caution should be exercised when administered to a nursing mother.
