Olanzapine is an atypical antipsychotic medication used to treat schizophrenia and bipolar disorder. It helps to restore the balance of certain natural substances in the brain.
Uses & Indications
Dosage
Adults
Schizophrenia: Initially 5-10 mg once daily, titrate to target dose of 10-15 mg/day. Bipolar mania: Initially 10-15 mg once daily.
Elderly
A lower starting dose of 5 mg/day is recommended for elderly patients, with careful titration.
Renal_impairment
No routine dose adjustment is required in renal impairment, but caution is advised.
How to Take
Take orally once daily, with or without food. Tablets can be swallowed whole with water.
Mechanism of Action
Olanzapine acts as an antagonist at multiple receptors including serotonin 5-HT2A, dopamine D1, D2, D3, D4, histamine H1, alpha1-adrenergic, and muscarinic M1, M2, M3, M4, M5 receptors. This multifaceted action contributes to its antipsychotic and mood-stabilizing effects.

Extensively metabolized in the liver, primarily via direct glucuronidation and cytochrome P450-mediated oxidation (CYP1A2 and CYP2D6).
