Pol 500 mg Tablet contains Paracetamol, a widely used medication for relieving pain and reducing fever.
Uses & Indications
Dosage
Adults
500 mg to 1000 mg (1-2 tablets) every 4-6 hours as needed. Maximum 4000 mg (8 tablets) in 24 hours.
Elderly
Generally same as adults, but caution with hepatic/renal impairment. Consult doctor.
Renal_impairment
Dosage interval may need to be extended (e.g., 6-8 hours for CrCl 10-50 mL/min, 8-12 hours for CrCl <10 mL/min). Consult doctor.
How to Take
Take orally with water, with or without food. Do not exceed the recommended dose or frequency.
Mechanism of Action
Paracetamol (acetaminophen) is thought to act primarily in the central nervous system, inhibiting prostaglandin synthesis through a mechanism not fully understood, but potentially involving selective inhibition of COX-3 or modulation of the endocannabinoid system. It has weak peripheral anti-inflammatory effects.
Pharmacokinetics
Onset
30-60 minutes for analgesia, similar for antipyresis.
Excretion
Excreted mainly in the urine, predominantly as glucuronide and sulfate conjugates. Less than 5% is excreted unchanged.
Half life
1-3 hours in adults, slightly longer in neonates and patients with hepatic or renal impairment.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract, with peak plasma concentrations occurring within 10-60 minutes after oral administration.
Metabolism
Primarily metabolized in the liver via glucuronidation and sulfation, and a small amount by cytochrome P450 to a highly reactive intermediate (N-acetyl-p-benzoquinone imine or NAPQI) which is rapidly detoxified by conjugation with glutathione.

- Hypersensitivity to paracetamol or any component of the formulation.
- Severe hepatic impairment or active liver disease.
Drug Interactions
Alcohol
Chronic alcohol consumption increases the risk of hepatotoxicity with paracetamol, even at therapeutic doses.
Warfarin
May increase the anticoagulant effect of warfarin, leading to an increased risk of bleeding, especially with prolonged use or higher doses. Regular monitoring of INR is recommended.
Other paracetamol-containing products
Increased risk of overdose and severe liver damage.
