Overview of the medicine
Preofloxin 500 mg Tablet is a broad-spectrum fluoroquinolone antibiotic used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
500 mg orally once daily for 7-14 days, depending on the infection type and severity.
Elderly
No specific dose adjustment is needed based on age alone, but renal function should be assessed.
Renal_impairment
For creatinine clearance 30-50 mL/min: 500 mg every 24 hours. For creatinine clearance <30 mL/min: 250 mg every 24 hours.
How to Take
Take the tablet orally with a glass of water, with or without food. Do not chew or crush the tablet.
Mechanism of Action
Preofloxin inhibits bacterial DNA gyrase and topoisomerase IV, essential enzymes involved in DNA replication, transcription, repair, and recombination, leading to bacterial cell death.

Onset
Peak plasma concentrations are achieved within 1-2 hours.
Excretion
Primarily excreted by the kidneys (both glomerular filtration and tubular secretion).
Half life
Approximately 6-8 hours.
Absorption
Rapidly and well absorbed from the gastrointestinal tract after oral administration. Bioavailability is approximately 70-80%.
Metabolism
Minimally metabolized in the liver; primarily excreted as unchanged drug.
Side Effects
Contraindications
- Hypersensitivity to preofloxin or any other fluoroquinolone.
- History of tendon disorders related to fluoroquinolone administration.
- Children and adolescents below 18 years of age (due to risk of arthropathy).
Drug Interactions
NSAIDs
May increase the risk of CNS stimulation and convulsive seizures.
Warfarin
May enhance the anticoagulant effect of warfarin. Monitor INR closely.
Corticosteroids
Increased risk of tendinitis and tendon rupture.
Antacids, Iron, Zinc, Sucralfate
May reduce preofloxin absorption. Administer preofloxin 2 hours before or 6 hours after these agents.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive treatment should be initiated. Gastric lavage and activated charcoal may be considered. Monitor ECG due to potential for QT prolongation. Maintain adequate hydration.
Pregnancy & Lactation
Preofloxin should not be used during pregnancy or lactation unless the potential benefit justifies the potential risk to the fetus or infant. Animal studies have shown adverse effects on cartilage development. Consult a doctor.
