Description
Overview of the medicine
Priocin 500 mg Tablet contains Clindamycin, a lincosamide antibiotic used to treat various serious bacterial infections including those of the skin, soft tissue, respiratory tract, gynecological organs, and intra-abdominal areas. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
For serious infections, 150-300 mg every 6 hours. For more severe infections, 300-450 mg every 6 hours. A 500 mg tablet might be prescribed for severe cases, often once or twice daily depending on infection and physician's assessment.
Elderly
No specific dosage adjustment is required based solely on age, but renal and hepatic function should be monitored.
Renal_impairment
Dose adjustment is generally not necessary in mild to moderate renal impairment. In severe renal impairment, consider dose reduction or increased dosing interval.
How to Take
Take the tablet orally with a full glass of water, with or without food, to prevent esophageal irritation. Do not lie down for at least 30 minutes after taking the medicine.
Mechanism of Action
Clindamycin inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, thereby preventing peptide chain elongation. This action leads to bacteriostatic effects.
Pharmacokinetics
Onset
Within 1 hour.
Excretion
Approximately 10% of the dose is excreted in urine and 4% in feces as active drug or metabolites. The remainder is eliminated as inactive metabolites.
Half life
Approximately 2-3 hours in adults with normal renal and hepatic function. May be prolonged in patients with severe hepatic or renal impairment.
Absorption
Rapidly and almost completely absorbed (about 90%) after oral administration. Peak plasma concentrations are achieved in about 45-60 minutes.
Metabolism
Extensively metabolized in the liver to active and inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to clindamycin, lincomycin, or any component of the formulation.
- History of C. difficile-associated diarrhea (CDAD) or other severe gastrointestinal disease.
- Infants younger than one month of age.
Drug Interactions
Erythromycin
Antagonistic effect; do not administer concurrently.
Kaolin-pectin antidiarrheals
May reduce the absorption of oral clindamycin.
Neuromuscular blocking agents
May enhance the neuromuscular blocking effects, leading to prolonged muscle paralysis.
Warfarin and other vitamin K antagonists
May increase INR and risk of bleeding.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, seek emergency medical attention. Treatment is primarily supportive; hemodialysis and peritoneal dialysis are not effective in removing clindamycin from the blood.
Pregnancy & Lactation
Pregnancy Category B. Studies in pregnant women have not shown an increased risk of fetal abnormalities. Use during pregnancy only if clearly needed. Clindamycin is excreted in breast milk; therefore, exercise caution when administering to a nursing mother. Monitor breastfed infants for diarrhea or rash.
