Description
Overview of the medicine
Prucon is a medication containing Prucalopride, used for the symptomatic treatment of chronic constipation in adults when laxatives fail to provide adequate relief. It works by stimulating colonic motility.
Uses & Indications
Dosage
Adults
1 mg once daily. If needed and tolerated, the dose can be increased to 2 mg once daily.
Elderly
Start with 1 mg once daily. Consider increasing to 2 mg if necessary and tolerated.
Renal_impairment
For severe renal impairment (CrCl < 30 mL/min), 1 mg once daily. No dosage adjustment needed for mild to moderate renal impairment.
How to Take
Prucon tablets can be taken with or without food, at any time of the day. Swallow the tablet whole with water.
Mechanism of Action
Prucalopride is a selective, high-affinity 5-HT4 receptor agonist that stimulates colonic motility, leading to increased bowel movements and relief from chronic constipation. It promotes propulsive motility in the colon.

Onset
Onset of action within 24 hours for some patients, full effect may take 1-2 weeks.
Excretion
Mainly excreted unchanged in urine (about 60-65%) and feces (about 25-30%).
Half life
Approximately 24-28 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentration reached in 2-3 hours. Bioavailability is >90%.
Metabolism
Minimal hepatic metabolism; primarily excreted unchanged.
Side Effects
Contraindications
- Hypersensitivity to prucalopride or any of the excipients.
- Intestinal obstruction, intestinal perforation, or other inflammatory bowel conditions (e.g., Crohn's disease, ulcerative colitis, toxic megacolon/megarectum).
- Severe renal impairment requiring dialysis.
Drug Interactions
Rifampicin
May decrease prucalopride plasma levels due to CYP3A4 induction.
Ketoconazole, Erythromycin, Clarithromycin
May increase prucalopride plasma levels due to CYP3A4 inhibition, but generally not clinically significant.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
