Description
Overview of the medicine
Prucalopride is a dihydrobenzofurancarboxamide derivative, a selective, high-affinity 5-HT4 receptor agonist that is used to treat chronic idiopathic constipation.
Uses & Indications
Dosage
Adults
2 mg once daily. For some patients, 1 mg once daily may be sufficient.
Elderly
Consider starting with 1 mg once daily, especially in patients with impaired renal function.
Renal_impairment
For severe renal impairment (CrCl < 30 mL/min), 1 mg once daily. No dosage adjustment needed for mild to moderate renal impairment.
How to Take
Take orally, with or without food, at any time of day.
Mechanism of Action
Prucalopride selectively stimulates 5-HT4 receptors in the gastrointestinal tract, promoting colonic motility and facilitating bowel movements. It enhances colonic peristalsis and defecation reflexes.

Contraindications

Drug Interactions
Erythromycin
No clinically significant interaction with erythromycin (another CYP3A4 inhibitor).
Ketoconazole
Co-administration with ketoconazole (a strong CYP3A4 inhibitor) increases prucalopride exposure; however, this is not considered clinically significant.
Oral contraceptives
No impact on the efficacy of oral contraceptives.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include headache, nausea, and diarrhea. Management is supportive and symptomatic.
