Description
Overview of the medicine
Respira 1 mg Syrup is an antihistamine used to relieve allergy symptoms such as sneezing, runny nose, itchy eyes, and hives. It works by blocking histamine, a natural substance that the body produces during an allergic reaction.
Uses & Indications
Dosage
Adults
Adults and adolescents 12 years and older: 5 mg (10 ml) once daily.
Elderly
No specific dosage adjustment needed, but caution with comorbidities.
Renal_impairment
Severe renal impairment: 5 mg (10 ml) every other day.
How to Take
Respira syrup can be taken with or without food. Use a measuring spoon or oral syringe for accurate dosing.
Mechanism of Action
Desloratadine, the active metabolite of loratadine, is a non-sedating, long-acting histamine antagonist with selective peripheral H1-receptor antagonist activity. It inhibits the release of histamine from mast cells and reduces inflammatory mediators.
Pharmacokinetics
Onset
Symptomatic relief usually occurs within 30-60 minutes.
Excretion
Eliminated equally in urine and feces, primarily as conjugated metabolites.
Half life
Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations in approximately 3 hours.
Metabolism
Extensively metabolized in the liver to 3-hydroxydesloratadine, an active metabolite, via hydroxylation followed by glucuronidation. CYP2D6 is involved.
Side Effects
Contraindications
- Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
- Severe kidney or liver disease (requires dose adjustment).
