Description
Overview of the medicine
Rufast 10 mg Tablet contains Rupatadine, a second-generation antihistamine, used to relieve symptoms of allergic rhinitis (such as sneezing, runny nose, itchy eyes, and nasal congestion) and chronic idiopathic urticaria (hives). It is a non-sedating antihistamine.
Uses & Indications
Dosage
Adults
10 mg once daily, taken with or without food.
Elderly
No specific dose adjustment is required, but use with caution due to potential decreased renal or hepatic function.
Renal_impairment
Use with caution. Data are limited for patients with severe renal impairment.

Use with caution. Data are limited for patients with severe hepatic impairment.
How to Take
Rufast 10 mg Tablet is for oral administration. It can be taken with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Rupatadine is a potent, long-acting, selective peripheral H1-receptor antagonist. It also has antagonistic activity against platelet-activating factor (PAF). These actions contribute to its antihistaminic and anti-inflammatory effects.

Onset
Onset of action typically occurs within 15-60 minutes.
Excretion
Mainly excreted as metabolites in urine (60.9%) and feces (34.6%).
Half life
The elimination half-life is approximately 5.9 hours.
Absorption
Rapidly absorbed after oral administration, with a Tmax of approximately 0.75-1 hour.
Metabolism
Extensively metabolized by the cytochrome P450 system (CYP3A4) into active and inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to rupatadine or any of the excipients.
- •Patients with severe renal or hepatic impairment (use with caution due to limited data).
Drug Interactions
Diltiazem
Moderate CYP3A4 inhibitor, may increase rupatadine levels. Use with caution.
Ketoconazole and Erythromycin
Potent CYP3A4 inhibitors, increase systemic exposure to rupatadine. Concomitant use should be with caution.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive treatment should be initiated. Activated charcoal may be considered. There is no specific antidote.
Pregnancy & Lactation
Pregnancy Category B. Limited data on pregnant women, use only if clearly needed and potential benefits outweigh risks. It is unknown whether rupatadine is excreted in human milk; a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
