Description
Overview of the medicine
Rupatadine is a non-sedating, long-acting antihistamine with selective peripheral H1-receptor antagonist activity. It is used to treat allergic rhinitis and urticaria in adults and adolescents (12 years and above).
Uses & Indications
Dosage
Adults
10 mg once daily.
Elderly
No special dosage adjustment is usually required, but caution should be exercised in patients with renal or hepatic impairment.
Renal_impairment
Caution, dose adjustment may be necessary in severe cases. Not recommended for patients with severe renal impairment without medical consultation.
How to Take
For oral use. Can be taken with or without food.
Mechanism of Action
Rupatadine acts as a selective peripheral H1-receptor antagonist. It also has antagonist activity against platelet-activating factor (PAF) and inhibits mast cell degranulation and release of inflammatory mediators.

Onset
Within 1 hour.
Excretion
Mainly via feces (60.9%) and urine (34.6%) as metabolites.
Half life
Approximately 5.9 hours (parent drug).
Absorption
Rapidly absorbed after oral administration. Tmax is approximately 0.75-1.5 hours.
Metabolism
Extensively metabolized by the liver via the cytochrome P450 (CYP3A4) pathway.

- Hypersensitivity to rupatadine or any excipients of the tablet.
Drug Interactions
Grapefruit Juice
May increase rupatadine exposure.
Ketoconazole and Erythromycin
Can increase systemic exposure to rupatadine due to CYP3A4 inhibition.
CNS Depressants (e.g., Alcohol)
May enhance central nervous system effects, leading to increased drowsiness.
