Overview of the medicine
Sigtil 100 mg tablet contains Sitagliptin, an oral antidiabetic drug used to improve blood sugar control in adults with type 2 diabetes mellitus. It belongs to a class of medicines called DPP-4 inhibitors.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg once daily. Can be taken with or without food.
Elderly
No dose adjustment required based on age; however, renal function should be assessed before initiating and periodically during treatment.
Renal_impairment
For moderate renal impairment (CrCl ≥30 to <50 mL/min): 50 mg once daily. For severe renal impairment (CrCl <30 mL/min) or end-stage renal disease requiring dialysis: 25 mg once daily.
How to Take
Take the tablet orally once daily, with or without food, as directed by your doctor.
Mechanism of Action
Sitagliptin selectively inhibits the enzyme dipeptidyl peptidase-4 (DPP-4), which is responsible for inactivating the incretin hormones GLP-1 and GIP. By inhibiting DPP-4, Sitagliptin increases the levels of active incretin hormones, leading to enhanced glucose-dependent insulin secretion and suppressed glucagon secretion from the pancreas.

Primarily excreted unchanged in the urine (approximately 79%).

Absorption

Limited metabolism, primarily by CYP3A4 and to a lesser extent by CYP2C8.

Digoxin
Increased plasma concentrations of digoxin were observed; monitor digoxin levels if co-administered.
