Description
Overview of the medicine
Sinacet 25 mg Oral Solution is an anti-inflammatory, analgesic, and antipyretic medicine used for the relief of pain, fever, and inflammation. It belongs to the class of non-steroidal anti-inflammatory drugs (NSAIDs).
Uses & Indications
Dosage
Adults
Typically 50-100 mg (10-20 mL) three times daily, or as directed by physician. Maximum daily dose 300 mg.
Elderly
Lower doses may be required; monitor for adverse effects.
Renal_impairment
Use with caution; dose adjustment may be necessary based on creatinine clearance.
How to Take
Administer orally. Can be taken with or without food. To minimize gastric irritation, it is often recommended to take it with food or milk.
Mechanism of Action
Sinacetin, the active ingredient, works by inhibiting the activity of cyclooxygenase (COX) enzymes, specifically COX-1 and COX-2. This leads to a reduction in the synthesis of prostaglandins, which are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Mainly excreted via urine as metabolites.
Half life
Approximately 2-4 hours.
Absorption
Rapidly and extensively absorbed from the gastrointestinal tract.
Metabolism
Primarily metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- Known hypersensitivity to sinacetin or other NSAIDs (e.g., aspirin-induced asthma)
- Active gastrointestinal bleeding or peptic ulcer disease
- Severe renal or hepatic impairment
- Third trimester of pregnancy
Drug Interactions
Lithium
Increased plasma lithium levels.
Methotrexate
Increased methotrexate toxicity.
ACE Inhibitors/ARBs
Increased risk of renal impairment and reduced antihypertensive effects.
Diuretics (e.g., Furosemide)
Reduced diuretic and antihypertensive effects.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store below 30°C in a cool, dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, lethargy, drowsiness, and occasionally more severe effects like gastrointestinal bleeding, acute renal failure, or respiratory depression. Management involves symptomatic and supportive treatment; gastric lavage or activated charcoal may be considered within an hour of ingestion.
Pregnancy & Lactation
Avoid use in the third trimester of pregnancy due to potential risk of premature closure of the ductus arteriosus and renal dysfunction in the fetus. Use with caution during lactation; small amounts may pass into breast milk.
