Description
Overview of the medicine
Singlin 0.5 mg Tablet is a non-benzodiazepine anxiolytic used for the short-term management of anxiety disorders.
Uses & Indications
Dosage
Adults
Typically 0.5 mg once daily, adjusted based on patient response and tolerability, up to a maximum of 2 mg daily in divided doses.
Elderly
Initial dose 0.25 mg once daily, increasing cautiously as needed. Lower doses may be sufficient due to reduced metabolism.
Renal_impairment
No specific dosage adjustment required for mild to moderate renal impairment. Use with caution in severe renal impairment.
How to Take
Take orally with water, with or without food. Do not crush or chew the tablet.
Mechanism of Action
Acts on specific serotonin receptors (e.g., 5-HT1A partial agonist) in the brain, reducing neuronal excitability and thereby exerting anxiolytic effects.
Pharmacokinetics
Onset
Anxiolytic effects typically begin within 30-60 minutes.
Excretion
Primarily excreted in the urine as metabolites, with a small portion in feces.
Half life
Approximately 6-8 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract, with peak plasma concentrations reached within 1-2 hours.
Metabolism
Extensively metabolized in the liver via CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to Singlin or any component of the tablet
- Severe respiratory insufficiency
- Acute narrow-angle glaucoma
Drug Interactions
CNS Depressants
Increased sedative effects when co-administered with other CNS depressants (e.g., alcohol, opioids, other sedatives).
CYP3A4 Inhibitors
Co-administration with potent CYP3A4 inhibitors (e.g., ketoconazole, erythromycin) may increase Singlin plasma concentrations, requiring dose adjustment.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include somnolence, confusion, reduced reflexes, and potentially respiratory depression. Treatment is supportive, including gastric lavage and activated charcoal if appropriate. Monitor vital signs.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended during breastfeeding as it may be excreted in breast milk.
