Description
Overview of the medicine
Sitagil 50 mg Tablet contains Sitagliptin, an oral anti-diabetic drug used to improve blood sugar control in adults with type 2 diabetes mellitus. It is used as an adjunct to diet and exercise.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg once daily, with or without food. For Sitagil 50 mg, it's typically used when titration or lower doses are required.
Elderly
No dosage adjustment is required based on age alone. Adjust dosage for renal impairment.
Renal_impairment
For moderate renal impairment (CrCl ≥30 to <50 mL/min), the dose is 50 mg once daily. For severe renal impairment (CrCl <30 mL/min) or ESRD requiring dialysis, the dose is 25 mg once daily.
How to Take
Take Sitagil 50 mg Tablet orally once daily, with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Sitagliptin is a dipeptidyl peptidase-4 (DPP-4) inhibitor. It works by inhibiting the enzyme DPP-4, which is responsible for inactivating incretin hormones (GLP-1 and GIP). By preserving active incretin levels, Sitagliptin increases insulin synthesis and release from pancreatic beta cells and decreases glucagon secretion from pancreatic alpha cells, both in a glucose-dependent manner. This leads to improved glycemic control.

Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached in 1 to 4 hours.

Limited metabolism, primarily by CYP3A4, with minor contribution from CYP2C8.
Side Effects
Contraindications
- Hypersensitivity to Sitagliptin or any component of the tablet.
- Patients with type 1 diabetes mellitus.
- Patients with diabetic ketoacidosis.
Drug Interactions
Digoxin
Sitagliptin has a minor effect on digoxin plasma concentrations; no dosage adjustment is generally needed for digoxin.
Cyclosporine
No clinically meaningful interaction observed.

Store in a cool and dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
In the event of an overdose, initiate appropriate supportive treatment as dictated by the patient’s clinical status. Hemodialysis removes approximately 13.5% of the dose over a 3- to 4-hour dialysis session.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed. It is not known whether Sitagliptin is excreted in human milk; caution should be exercised when administered to a nursing woman.
