Description
Overview of the medicine
Solider 10 mg Tablet is used to treat symptoms of overactive bladder, such as frequent urination, urgent need to urinate, and inability to control urination (urge incontinence).
Uses & Indications
Dosage
Adults
Recommended starting dose is 5 mg once daily. May be increased to 10 mg once daily based on response and tolerability.
Elderly
No dose adjustment is generally required, but caution is advised due to potential for increased sensitivity.
Renal_impairment
For severe renal impairment (CrCl < 30 mL/min), maximum dose is 5 mg once daily. Use with caution.
How to Take
Take orally once daily with water, with or without food. Swallow the tablet whole, do not crush or chew.
Mechanism of Action
Solifenacin is a competitive muscarinic receptor antagonist. It selectively blocks M3 muscarinic receptors primarily in the bladder, reducing detrusor muscle contractions and thereby decreasing urinary frequency, urgency, and incontinence.
Pharmacokinetics
Onset
Within a few hours for initial effects; full therapeutic effect typically within 2-4 weeks.
Excretion
Primarily excreted in urine (approximately 69%), with a smaller portion in feces (approximately 23%).
Half life
Approximately 45-68 hours.
Absorption
Well absorbed from the gastrointestinal tract, with absolute bioavailability of about 90%.
Metabolism
Extensively metabolized in the liver, primarily by CYP3A4, to several inactive and one active metabolite.
Side Effects
Contraindications
- Urinary retention
- Gastric retention
- Uncontrolled narrow-angle glaucoma
- Severe liver impairment
- Hypersensitivity to solifenacin or any excipients
Drug Interactions
Other anticholinergic agents
May potentiate anticholinergic effects.
CYP3A4 inducers (e.g., rifampicin)
May decrease solifenacin efficacy.
Strong CYP3A4 inhibitors (e.g., ketoconazole)
May increase solifenacin exposure; limit solifenacin dose to 5 mg/day.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms may include severe anticholinergic effects like dry mouth, constipation, blurred vision, tachycardia, urinary retention, and drowsiness. Treatment is supportive, and gastric lavage may be considered. Physostigmine may be used in severe cases.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit justifies potential risk to the fetus. It is not known whether solifenacin is excreted in human milk; caution should be exercised when administered to a nursing woman.
