Overview of the medicine
Staflu 250 mg Capsule contains Flucloxacillin, a penicillinase-resistant penicillin antibiotic used to treat various bacterial infections, especially those caused by penicillinase-producing staphylococci.
Uses & Indications
Dosage
Adults
Usual dose is 250-500 mg every 6 hours, preferably 30-60 minutes before meals. For severe infections, the dose may be increased.
Elderly
No specific dose reduction is usually required unless there is significant renal impairment. Use with caution and monitor renal function.
Renal_impairment
In severe renal impairment (creatinine clearance <10 mL/min), the dose should be reduced or the dosing interval prolonged (e.g., 250 mg every 8 or 12 hours) to avoid accumulation.
How to Take
Take orally, preferably 30-60 minutes before meals, with a glass of water. Do not chew or crush the capsule.
Mechanism of Action
Flucloxacillin acts by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located in the bacterial cell wall, which prevents the cross-linking of peptidoglycan polymers, leading to cell lysis and bacterial death.

Primarily excreted unchanged in the urine, with some excretion via bile.

Drug Interactions
Warfarin
May enhance the anticoagulant effect of warfarin. Close monitoring of INR and dose adjustment of warfarin may be necessary.
Probenecid
Probenecid delays the renal excretion of flucloxacillin, leading to increased and prolonged plasma concentrations.
Methotrexate
Flucloxacillin may reduce the renal excretion of methotrexate, potentially increasing its toxicity.
