Overview of the medicine
Tioconazole 1 gm vaginal tablet is an antifungal medication used for the local treatment of vulvovaginal candidiasis (yeast infections) caused by Candida species.
Uses & Indications
Dosage
Adults
Insert one 1 gm vaginal tablet deep into the vagina as a single dose, preferably at bedtime.
Elderly
No specific dosage adjustment is required for elderly patients; use as for adults.
Renal_impairment
No dosage adjustment is necessary for patients with renal impairment due to minimal systemic absorption.
How to Take
The vaginal tablet should be inserted as deeply as possible into the vagina, preferably at bedtime. An applicator may be provided to aid insertion.
Mechanism of Action
Tioconazole works by altering the permeability of the fungal cell membrane. It inhibits the biosynthesis of ergosterol, a vital component of fungal cell membranes, leading to increased permeability, leakage of cellular contents, and eventual death of fungal cells.

Onset
Symptom improvement usually begins within 2-3 days following administration.
Excretion
Systemically absorbed Tioconazole is primarily excreted via urine and feces; however, the amount is negligible due to minimal absorption.
Half life
Systemic half-life is not clinically relevant due to very low systemic absorption; local action is predominant.
Absorption
Minimal systemic absorption occurs after vaginal administration. Less than 10% of the dose is absorbed systemically.
Metabolism
Minimal systemic metabolism occurs due to low absorption.

Contraindications
- Hypersensitivity to Tioconazole or any other imidazole antifungal agents.

Drug Interactions
Warfarin
Although systemic absorption is minimal, there have been rare reports of increased anticoagulant effect when vaginal imidazole antifungals are used concomitantly with oral anticoagulants like warfarin. Monitor INR if co-administration is necessary.

Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Overdose with topical vaginal Tioconazole is highly unlikely due to minimal systemic absorption. In the event of accidental oral ingestion, general supportive measures should be taken.
