Description
Overview of the medicine
Tinidazole is an antiprotozoal and antibacterial agent used to treat various infections caused by susceptible protozoa and anaerobic bacteria.
Uses & Indications
Dosage
Adults
Dosage for Tinidazole typically ranges from 500 mg to 2 g, depending on the indication. For 50 mg formulation, specific dosage should be strictly followed as prescribed by a physician, as this strength is unusual for standard indications.
Elderly
No specific dosage adjustment is generally required, but caution is advised in patients with impaired renal or hepatic function.
Renal_impairment
In severe renal impairment (creatinine clearance <10 mL/min), the dosage interval may need to be prolonged, or the dose reduced. Consultation with a physician is recommended.
How to Take
Tinimet tablets should be taken orally, preferably with or after food, to minimize gastrointestinal discomfort. Do not chew or crush the tablet; swallow it whole.
Mechanism of Action
Tinidazole acts as a nitroimidazole derivative. It enters the microbial cell, where its nitro group is reduced, forming reactive intermediates that damage DNA and inhibit nucleic acid synthesis, leading to cell death.
Pharmacokinetics
Onset
Within 1-2 hours for systemic effects.
Excretion
Excreted by both renal (approximately 20-25% as unchanged drug) and fecal routes.
Half life
Approximately 12-14 hours, allowing for once-daily or single-dose regimens.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are achieved within 2 hours.
Metabolism
Primarily metabolized in the liver by oxidation, hydroxylation, and conjugation.
Side Effects
Contraindications
- •Hypersensitivity to tinidazole or other nitroimidazole derivatives.
- •First trimester of pregnancy.
- •Patients with active neurological disorders or a history of blood dyscrasias.
Drug Interactions
Alcohol
Disulfiram-like reaction (abdominal cramps, nausea, vomiting, headache, flushing) when consumed during or within 72 hours of treatment.
Cimetidine
May reduce the hepatic metabolism of tinidazole, potentially increasing its plasma concentrations.
Warfarin and other oral anticoagulants
May potentiate the anticoagulant effect, requiring close monitoring of INR and dose adjustment.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, ataxia, and neurological symptoms. Treatment is symptomatic and supportive. There is no specific antidote for tinidazole overdose. Hemodialysis may be useful in removing the drug.
Pregnancy & Lactation
Contraindicated during the first trimester of pregnancy. Use with caution in the second and third trimesters only if clearly needed. Tinidazole is excreted into breast milk; breastfeeding should be avoided during treatment and for at least 72 hours after the last dose.
