Topup 10 mg Tablet contains Cholecalciferol (Vitamin D3), a fat-soluble vitamin essential for calcium and phosphate absorption, bone health, and immune function. This high-dose formulation is typically used to treat severe vitamin D deficiency.
Uses & Indications
Dosage
Adults
For severe deficiency, 10 mg (400,000 IU) once every 3-4 months, or as directed by physician, usually followed by lower maintenance doses. Individual dosing varies.
Elderly
Same as adults, but monitoring for renal function and calcium levels may be more frequent.
Renal_impairment
Use with caution and monitor calcium and phosphate levels; dose adjustment may be necessary. Active vitamin D metabolites (calcitriol) may be preferred in severe renal impairment.
How to Take
Oral, swallow whole with water. Can be taken with food for better absorption.
Mechanism of Action
Cholecalciferol is metabolized in the liver to 25-hydroxycholecalciferol (calcidiol) and then in the kidneys to 1,25-dihydroxycholecalciferol (calcitriol), the active form of vitamin D. Calcitriol acts as a hormone to regulate calcium and phosphate levels by promoting absorption from the gut, reabsorption in the kidneys, and mobilization from bone.

Excretion
Mainly excreted in the bile and feces, with a small amount in urine.
Half life
Circulating half-life of 25-hydroxyvitamin D is about 2-3 weeks; active form 1,25-dihydroxyvitamin D has a half-life of 4-6 hours.

Readily absorbed from the gastrointestinal tract in the presence of bile salts.
Metabolism
