Description
Overview of the medicine
Triject 1 gm Injection is an antibiotic belonging to the cephalosporin class. It is used to treat a variety of bacterial infections in different parts of the body, including severe infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Typical adult dosage is 1-2 gm once daily (OD) or in two divided doses for 4-14 days depending on the infection. For severe infections, up to 4 gm daily.
Elderly
No specific dose adjustment is generally required for elderly patients unless there is severe renal or hepatic impairment.
Renal_impairment
Dose adjustment may be necessary in severe renal impairment, especially with concomitant hepatic impairment. No dose adjustment needed for mild to moderate impairment.
How to Take
Administered intravenously (IV) or intramuscularly (IM) by a healthcare professional. IV administration should be slow (over 30 minutes) to avoid vein irritation.
Mechanism of Action
Ceftriaxone inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Rapid, within 30 minutes for IV administration
Excretion
Approximately 50-60% excreted unchanged in urine, and 40-50% excreted via bile into feces.
Half life
Approximately 5.8 to 8.7 hours
Absorption
Rapidly and completely absorbed after intramuscular (IM) administration; high bioavailability.
Metabolism
Limited metabolism; primarily in the liver (small amount to inactive metabolites)
Side Effects
Contraindications
- Hypersensitivity to ceftriaxone or any other cephalosporin or penicillin
- Neonates (especially premature) with hyperbilirubinemia
- Neonates receiving calcium-containing IV solutions
Drug Interactions
Aminoglycosides
Potential for synergistic effect against certain bacteria, but no known pharmacokinetic interaction.
Oral anticoagulants (e.g., Warfarin)
Increased risk of bleeding due to vitamin K antagonism; monitor INR.
Calcium-containing solutions (e.g., Ringer's, Hartmann's)
Formation of precipitates, especially in neonates. Fatal reactions reported.
Storage
Store below 30°C, protected from light and moisture. Keep out of reach of children. Reconstituted solution should be used immediately or stored as per manufacturer's guidelines.
Overdose
Overdose symptoms may include nausea, vomiting, diarrhea, and central nervous system effects (e.g., convulsions). Treatment is supportive and symptomatic; hemodialysis or peritoneal dialysis is not effective.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed. Excreted in low concentrations in breast milk; generally considered compatible with breastfeeding, but monitor infant for adverse effects.
