Trilock 10 mg Tablet contains Atorvastatin, a medication used to lower high cholesterol levels and reduce the risk of heart disease, stroke, and other cardiovascular problems. It belongs to a group of medicines called statins.
Uses & Indications
Dosage
Adults
Initial dose is usually 10 mg once daily. The dose can be adjusted by a doctor based on LDL-C goals and patient response, up to a maximum of 80 mg daily.
Elderly
No specific dose adjustment is required based on age. Follow adult dosage recommendations.
Renal_impairment
No dosage adjustment is necessary for patients with renal impairment.
How to Take
Take Trilock 10 mg Tablet orally once daily, with or without food, at any time of the day. It is usually taken at the same time each day for consistency.
Mechanism of Action
Atorvastatin works by inhibiting HMG-CoA reductase, an enzyme involved in cholesterol production in the liver. This reduces the amount of cholesterol the body makes, particularly LDL (bad) cholesterol, and increases the liver's ability to remove LDL cholesterol from the blood.
Pharmacokinetics
Onset
Significant lipid-lowering effects typically observed within 2 weeks, with maximal response usually achieved by 4 weeks.
Excretion
Primarily excreted in the bile following hepatic and/or extrahepatic metabolism. Less than 2% is recovered in the urine.
Half life
Approximately 14 hours (for parent drug and active metabolites), but inhibition of HMG-CoA reductase activity persists for 20-30 hours due to active metabolites.
Absorption
Rapidly absorbed from the gastrointestinal tract; absolute bioavailability is approximately 12%. Peak plasma concentrations achieved within 1 to 2 hours.
Metabolism
Extensively metabolized by cytochrome P450 3A4 (CYP3A4) in the liver to active ortho- and parahydroxylated metabolites and various beta-oxidation products.

Contraindications
- Active liver disease or unexplained persistent elevations of serum transaminases
- Pregnancy and lactation
- Hypersensitivity to Atorvastatin or any component of the formulation
Drug Interactions
Warfarin
Potentiation of anticoagulant effect, requiring careful INR monitoring.
Gemfibrozil and other fibrates
Increased risk of myopathy when co-administered.
Clarithromycin, Erythromycin, Itraconazole
Increased plasma concentrations of atorvastatin, increasing risk of side effects.
