Description
Overview of the medicine
Tigecycline is a glycylcycline antibiotic indicated for the treatment of complicated skin and skin structure infections (cSSSI), complicated intra-abdominal infections (cIAI), and community-acquired bacterial pneumonia (CABP) caused by susceptible microorganisms.
Uses & Indications
Dosage
Adults
Initial dose: 100 mg intravenously, followed by 50 mg intravenously every 12 hours. Administered over 30 to 60 minutes.
Elderly
No dose adjustment is necessary for elderly patients.
Renal_impairment
No dose adjustment is necessary for patients with renal impairment or those undergoing hemodialysis.
How to Take
Tygacil is administered by intravenous infusion over 30 to 60 minutes. It must be reconstituted and then diluted before use.
Mechanism of Action
Tigecycline inhibits protein synthesis in bacteria by binding to the 30S ribosomal subunit, preventing the entry of aminoacyl-tRNA molecules into the A site of the ribosome. This action is similar to tetracyclines but with an enhanced binding affinity.

Side Effects
Contraindications
- •Hypersensitivity to tigecycline or any component of the formulation.
- •Known hypersensitivity to tetracycline class antibiotics (due to potential cross-sensitivity).
Drug Interactions
Warfarin
Concomitant use with warfarin may lead to an increase in prothrombin time (PT) or International Normalized Ratio (INR). Monitor coagulation parameters closely.
Oral Contraceptives
Tigecycline may reduce the effectiveness of oral contraceptives; advise patients to use an alternative or additional method of contraception.
Storage
Store unopened vials at 20-25°C (68-77°F). Protect from light. Do not freeze reconstituted solution.
Overdose
In the event of an overdose, discontinue Tygacil and provide supportive treatment. Tigecycline is not significantly removed by hemodialysis.
Pregnancy & Lactation
Pregnancy Category D. Tigecycline may cause permanent tooth discoloration and inhibit bone growth if used during pregnancy. It is excreted into breast milk; caution should be exercised when administered to a nursing woman.
