Valarux 500 mg Tablet is an antiviral medicine used to treat infections caused by herpes viruses, including cold sores, genital herpes, and shingles.
Uses & Indications
Dosage
Adults
Herpes Zoster: 1000 mg three times daily for 7 days. Genital Herpes (initial episode): 1000 mg twice daily for 7-10 days. Cold Sores: 2000 mg twice daily for 1 day.
Elderly
No dose adjustment is needed unless there is significant renal impairment. Monitor renal function.
Renal_impairment
Dosage adjustment is necessary based on creatinine clearance. For example, CrCl 30-49 mL/min: 1000 mg twice daily for shingles.
How to Take
Take Valarux 500 mg Tablet orally with or without food. It is best to start treatment as soon as possible after the first symptoms appear.
Mechanism of Action
Valarux (Valaclovir Hydrochloride) is a prodrug that is rapidly converted to acyclovir. Acyclovir inhibits viral DNA polymerase, preventing viral replication.
Pharmacokinetics
Onset
Peak plasma concentrations of acyclovir are reached within 1-2 hours.
Excretion
Primarily excreted unchanged in the urine as acyclovir.
Half life
The plasma elimination half-life of acyclovir is approximately 2.5-3.3 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract and almost completely converted to acyclovir and L-valine.
Metabolism
Converted to acyclovir by first-pass intestinal and hepatic metabolism.
Side Effects
Contraindications
- •Hypersensitivity to valaclovir, acyclovir, or any component of the formulation.
- •Severe immunocompromised patients in certain conditions.
Drug Interactions
Cimetidine
May slightly increase acyclovir levels.
Probenecid
Increases acyclovir levels, requiring dose adjustment.
Nephrotoxic drugs (e.g., NSAIDs, Cyclosporine)
May increase the risk of renal dysfunction; monitor renal function closely.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include acute renal failure and neurological symptoms (confusion, hallucinations, seizures). Treatment is symptomatic and supportive, including hemodialysis for severe cases.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Acyclovir is excreted in breast milk; caution is advised when administered to a nursing mother.
