Vanprox 40 mg Suspension contains Pantoprazole, a proton pump inhibitor used to reduce stomach acid production. It is prescribed for conditions like acid reflux, heartburn, ulcers, and other gastric acid-related disorders.
Uses & Indications
Dosage
Adults
For GERD/Erosive Esophagitis: 40 mg once daily for up to 8 weeks. For Zollinger-Ellison Syndrome: Dosage varies, usually starting with 80 mg once daily, adjusted based on acid output.
Elderly
No specific dose adjustment is generally required, but caution is advised in patients with severe hepatic impairment.
Renal_impairment
No dose adjustment is necessary for patients with renal impairment.
How to Take
Take the suspension orally, usually once daily, at least 30 minutes before a meal. Shake well before use. Do not crush or chew if it's a different form like tablet.
Mechanism of Action
Pantoprazole irreversibly inhibits the H+/K+-ATPase (proton pump) in the parietal cells of the stomach, thereby blocking the final step of gastric acid secretion.

Onset
Within 2.5 hours for acid suppression.
Excretion
Mainly excreted renally (approximately 71%) and a smaller amount (approximately 18%) via biliary excretion.
Half life
Approximately 1 hour, but the duration of acid suppression is much longer due to irreversible binding.
Absorption
Rapidly absorbed after oral administration; bioavailability is about 77%. Food may delay absorption but does not affect overall bioavailability.
Metabolism
Extensively metabolized in the liver via the cytochrome P450 (CYP) system, primarily by CYP2C19 and CYP3A4.

Contraindications
- Known hypersensitivity to Pantoprazole or any component of the formulation.
- Hypersensitivity to other substituted benzimidazoles.
Drug Interactions
Clopidogrel
Reduced effectiveness of Clopidogrel (antiplatelet agent).
Methotrexate
May increase Methotrexate levels, especially with high-dose Methotrexate.
Iron Salts, Vitamin B12
Decreased absorption due to reduced gastric acidity.
Ketoconazole, Itraconazole, Erlotinib
Absorption may be decreased due to increased gastric pH.
