Description
Overview of the medicine
Vardena 10 mg Tablet contains Vardenafil, used to treat erectile dysfunction (impotence) in men. It helps men to achieve and maintain an erection suitable for sexual activity by increasing blood flow to the penis when sexually stimulated.
Uses & Indications
Dosage
Adults
The recommended starting dose is 10 mg, taken orally as needed, approximately 25 to 60 minutes before sexual activity. The dose may be increased to 20 mg or decreased to 5 mg based on efficacy and tolerability. Maximum dose is one tablet per day.
Elderly
For patients ≥65 years, a starting dose of 5 mg is recommended.
Renal_impairment
No dose adjustment is required for patients with mild to moderate renal impairment. Not studied in severe renal impairment.
How to Take
Take Vardena 10 mg tablet orally, with or without food. Avoid high-fat meals as they may delay the effect. Do not take more than one tablet within a 24-hour period.
Mechanism of Action
Vardenafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5). When sexual stimulation causes local release of nitric oxide, PDE5 inhibition by vardenafil increases cGMP levels in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood into the penis.
Pharmacokinetics
Onset
Approximately 15-30 minutes.
Excretion
Excreted predominantly in the feces (91-95%) and to a lesser extent in the urine (2-6%).
Half life
The mean terminal half-life is approximately 4-5 hours.
Absorption
Rapidly absorbed. Peak plasma concentrations are reached within 30 minutes to 2 hours (median 60 minutes). Absolute bioavailability is approximately 15%. High-fat meals may delay absorption.
Metabolism
Primarily metabolized in the liver via cytochrome P450 (CYP3A4) and, to a lesser extent, CYP3A5 and CYP2C9.
Side Effects
Contraindications
- Concomitant use with nitrates or nitric oxide donors (e.g., nitroglycerin) due to potential for severe hypotension.
- Known hypersensitivity to Vardenafil or any component of the tablet.
- Concomitant use with alpha-blockers in certain patients due to increased risk of symptomatic hypotension.
Drug Interactions

Contraindicated due to potential for severe, life-threatening hypotensive effects.
Alpha-blockers
May cause symptomatic hypotension; dose adjustment of Vardenafil and/or careful monitoring required.
CYP3A4 Inhibitors (e.g., Ketoconazole, Ritonavir, Erythromycin)
Can significantly increase Vardenafil plasma levels, requiring dose reduction of Vardenafil.

In cases of overdose, standard supportive measures should be adopted as required. Renal dialysis is not expected to accelerate clearance, as vardenafil is highly bound to plasma proteins.
