Vardena 20 mg Tablet contains Vardenafil, a phosphodiesterase-5 (PDE5) inhibitor, used for the treatment of erectile dysfunction (impotence) in men.
Uses & Indications
Dosage
Adults
The usual recommended dose is 10 mg taken orally approximately 25 to 60 minutes before sexual activity. If 10 mg is not effective, the dose may be increased to 20 mg. For Vardena 20 mg tablet, patients should follow their doctor's advice, which may involve taking the full 20 mg or a half (10 mg) depending on individual needs. Maximum one dose per 24 hours.
Elderly
A starting dose of 5 mg is recommended for elderly patients (≥65 years).
Renal_impairment
No dosage adjustment is needed for mild to moderate renal impairment. Not studied in severe renal impairment.
How to Take
Take orally with or without food, approximately 25 to 60 minutes before sexual activity. Do not exceed one dose per day.
Mechanism of Action
Vardenafil inhibits phosphodiesterase-5 (PDE5), an enzyme responsible for the degradation of cyclic guanosine monophosphate (cGMP) in the corpus cavernosum. By inhibiting PDE5, Vardenafil increases cGMP levels, leading to smooth muscle relaxation and increased blood flow into the penis, resulting in an erection in response to sexual stimulation.

Contraindications

- Concomitant use with guanylate cyclase stimulators, such as riociguat.

Potentiation of hypotensive effects, potentially life-threatening. Absolutely contraindicated.
Riociguat
Contraindicated due to risk of symptomatic hypotension.
Alpha-blockers
May lead to symptomatic hypotension, especially with initial co-administration.
CYP3A4 inhibitors
Increase vardenafil plasma levels, increasing risk of side effects.

Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

Symptoms include severe back pain, muscle pain, visual disturbances. Management is supportive, as vardenafil is highly protein-bound and not expected to be dialyzable.
