Description
Overview of the medicine
Ventoxen 100 mg Tablet is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, swelling, and inflammation associated with various conditions such as osteoarthritis, rheumatoid arthritis, and acute pain.
Uses & Indications
Dosage
Adults
Osteoarthritis: 100 mg once daily. Rheumatoid Arthritis: 100 mg twice daily. Acute Pain/Dysmenorrhea: 100-200 mg once daily, or 100 mg twice daily as needed. Max 200 mg/day.
Elderly
No specific dose adjustment is generally required, but caution is advised due to potential age-related decrease in renal function.
Renal_impairment
Mild to moderate impairment: No dose adjustment. Severe impairment: Use with caution; consider lower doses.
How to Take
Ventoxen 100 mg Tablet should be taken orally, with or without food. Taking with food may reduce the risk of gastrointestinal upset. Swallow the tablet whole with a glass of water; do not crush, chew, or break it.
Mechanism of Action
Ventoxen selectively inhibits cyclooxygenase-2 (COX-2), an enzyme responsible for the synthesis of prostaglandins, which are mediators of pain and inflammation. By inhibiting COX-2, Ventoxen reduces the production of these inflammatory mediators, thereby alleviating pain and inflammation with a potentially lower risk of gastrointestinal side effects compared to non-selective NSAIDs.
Pharmacokinetics
Onset
Analgesic effect observed within 30-60 minutes.
Excretion
Excreted primarily via urine (approximately 50-60%) and feces (approximately 30-40%) as metabolites.
Half life
Approximately 8-12 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract, achieving peak plasma concentrations within 2-4 hours after oral administration. Bioavailability is approximately 80-90%.
Metabolism
Primarily metabolized in the liver by cytochrome P450 enzymes (CYP2C9) to inactive metabolites.
Side Effects
Contraindications
- Known hypersensitivity to ventoxen or any component of the formulation.
- History of asthma, urticaria, or other allergic-type reactions after taking aspirin or other NSAIDs.
- Active gastrointestinal bleeding or ulceration.
- Severe renal or hepatic impairment.
- Severe heart failure.
- Third trimester of pregnancy.
- Coronary artery bypass graft (CABG) surgery.
Drug Interactions
Lithium
May increase plasma lithium levels, leading to toxicity.
Diuretics
May reduce natriuretic effect of loop and thiazide diuretics.
Methotrexate
May increase methotrexate plasma levels, potentially leading to toxicity.
SSRIs and SNRIs
Increased risk of gastrointestinal bleeding.
Warfarin and other anticoagulants
Increased risk of bleeding.
ACE Inhibitors and Angiotensin Receptor Blockers (ARBs)
May reduce antihypertensive effect and increase risk of renal impairment.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, lethargy, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Rare symptoms include hypertension, acute renal failure, respiratory depression, and coma. Management is symptomatic and supportive. There is no specific antidote. Gastric lavage or activated charcoal may be considered if ingestion is recent.
Pregnancy & Lactation
Pregnancy Category C (first and second trimesters); Category D (third trimester). Avoid in late pregnancy due to potential for premature closure of the fetal ductus arteriosus. Use during lactation only if the potential benefit outweighs the potential risk to the infant. Consult a doctor.
