Description
Overview of the medicine
Ventus 10 mg Tablet contains Loratadine, a non-drowsy antihistamine used to relieve symptoms associated with allergic rhinitis (hay fever) and chronic idiopathic urticaria (hives). It helps alleviate sneezing, runny nose, itchy or watery eyes, and itchy skin rashes.
Uses & Indications
Dosage
Adults
10 mg once daily.
Elderly
No dosage adjustment necessary.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), initial dose of 10 mg every other day is recommended.
How to Take
Ventus 10 mg tablet should be taken orally, with or without food. It can be swallowed whole with water.
Mechanism of Action
Loratadine is a tricyclic antihistamine with selective peripheral H1-receptor antagonist activity. It prevents histamine from binding to H1 receptors, thereby blocking the symptoms associated with the allergic response, such as itching, sneezing, and vasodilation, with minimal sedative and anticholinergic effects.

Rapidly and well absorbed from the gastrointestinal tract. Peak plasma concentrations are reached in 1-1.5 hours.
Metabolism
Extensively metabolized in the liver by cytochrome P450 enzymes (CYP3A4 and CYP2D6) to its major active metabolite, descarboethoxyloratadine.

Alcohol
No potentiation of alcohol effects has been found.
Cimetidine
May increase plasma concentrations of loratadine and its active metabolite.
Erythromycin

Ketoconazole

Symptoms of overdose include somnolence, tachycardia, and headache. Management should be symptomatic and supportive. Gastric lavage and activated charcoal may be considered. Loratadine is not effectively removed by hemodialysis.

Pregnancy Category B. Studies in animals have not shown a risk to the fetus, but there are no adequate and well-controlled studies in pregnant women. Should be used during pregnancy only if clearly needed. Loratadine and its active metabolite are excreted in breast milk; therefore, caution should be exercised when administered to a nursing woman.
