Mechanism of Action
Paracetamol acts centrally to produce analgesia and antipyresis. Propyphenazone is a pyrazolone derivative with analgesic, antipyretic, and anti-inflammatory effects, primarily by inhibiting prostaglandin synthesis. Caffeine is a central nervous system stimulant that enhances the analgesic effects of paracetamol and propyphenazone and helps to alleviate fatigue.
Pharmacokinetics

Onset of action typically within 30-60 minutes.
Excretion
Mainly excreted by the kidneys as metabolites and unchanged drug.
Half life
Paracetamol: 1-4 hours; Propyphenazone: 2-3 hours; Caffeine: 3-7 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract. Peak plasma levels for paracetamol are usually reached within 30-60 minutes, propyphenazone within 1-2 hours, and caffeine within 30-60 minutes.
Metabolism
Primarily metabolized in the liver, mainly by conjugation (paracetamol, propyphenazone) and demethylation/oxidation (caffeine).
