Overview of the medicine
Zoledronic acid is a bisphosphonate medication used to treat osteoporosis, Paget's disease of bone, hypercalcemia of malignancy, and bone metastases from solid tumors. It works by slowing down bone loss and strengthening bones.
Uses & Indications
Dosage
Adults
Osteoporosis: 5 mg IV infusion once yearly. Paget's disease: 5 mg IV infusion once. Hypercalcemia of Malignancy: 4 mg IV infusion single dose. Bone metastases/skeletal events: 4 mg IV infusion every 3-4 weeks.
Elderly
No specific dose adjustment required based on age alone, but renal function should be monitored.
Renal_impairment
Dose adjustment or contraindication may apply for severe renal impairment (creatinine clearance < 35 mL/min).
How to Take
Administered as an intravenous infusion over at least 15-30 minutes, ensuring adequate hydration before and after infusion.
Mechanism of Action
Zoledronic acid inhibits osteoclast-mediated bone resorption. It binds to hydroxyapatite crystals in bone and is then internalized by osteoclasts, where it interferes with their mevalonate pathway, leading to osteoclast apoptosis and reduced bone turnover.
