Description
Overview of the medicine
Almoxil 500 mg capsule contains Amoxicillin, a penicillin-type antibiotic used to treat a wide range of bacterial infections.
Uses & Indications
Dosage
Adults
250 mg to 500 mg every 8 hours, or 875 mg every 12 hours, depending on the severity and type of infection. Max 4g/day.
Elderly
No specific dose adjustment generally needed unless severe renal impairment is present. Monitor renal function.
Renal_impairment
Dose reduction or increased dosing interval required based on creatinine clearance. For CrCl <30 mL/min, dosage adjustment is necessary.
How to Take
Take orally with or without food. Capsules should be swallowed whole with water.
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Primarily excreted unchanged by the kidneys (glomerular filtration and tubular secretion).
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract; bioavailability is approximately 70-90% and is not significantly affected by food.
Metabolism
Partially metabolized (about 10-25%) to inactive penicilloic acid.
Side Effects
Contraindications
- •Known hypersensitivity to penicillins or any component of the formulation.
- •History of severe hypersensitivity reaction (e.g., anaphylaxis) to any other beta-lactam agent (e.g., cephalosporins, carbapenems).
Drug Interactions
Allopurinol
Increased incidence of rash when co-administered with amoxicillin.
Methotrexate
Amoxicillin may decrease renal tubular secretion of methotrexate, potentially increasing its toxicity.
Oral contraceptives
May reduce the efficacy of oral contraceptives in some cases, advise alternative contraception.
Warfarin and other oral anticoagulants
May prolong prothrombin time (INR); caution advised, monitor INR.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include gastrointestinal effects (nausea, vomiting, diarrhea) and disturbances in fluid and electrolyte balance. Renal impairment can lead to amoxicillin crystalluria. Treatment is supportive and symptomatic.
Pregnancy & Lactation
Amoxicillin is generally considered safe for use during pregnancy (Category B) and lactation. Consult a doctor before use.
