Description
Overview of the medicine
Amoxicillin is a penicillin-type antibiotic used to treat a wide variety of bacterial infections. It works by stopping the growth of bacteria. This antibiotic treats only bacterial infections. It will not work for viral infections (e.g., common cold, flu).
Uses & Indications
Dosage
Adults
General dosage: 250 mg to 500 mg every 8 hours, or 750 mg to 1 g twice daily, depending on the severity and type of infection. Higher doses may be used for severe infections.
Elderly
Similar to adults, but dosage adjustment may be necessary if renal function is impaired. Consult a physician.
Renal_impairment
Dosage reduction is required. For creatinine clearance less than 30 mL/min, the dose interval should be increased or the dose reduced. Consult a physician for specific recommendations.
How to Take
Take Amovix tablets orally with a glass of water, with or without food. It is recommended to take it at evenly spaced intervals. Do not crush or chew the tablet; swallow it whole.
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis by binding to one or more of the penicillin-binding proteins (PBPs) which in turn inhibits the final transpeptidation step of peptidoglycan synthesis in the bacterial cell wall, thus inhibiting cell wall biosynthesis. This leads to the lysis and death of bacterial cells.
Pharmacokinetics
Onset
Clinical effects usually seen within a few hours to days, depending on the infection type and severity.
Excretion
Primarily excreted unchanged in the urine by active tubular secretion and glomerular filtration. Renal clearance accounts for 60-80% of total clearance.
Half life
Approximately 1 to 1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, even in the presence of food. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Approximately 10-25% of an oral dose is metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- Known hypersensitivity to amoxicillin, any other penicillin, or to any of the excipients.
- History of severe immediate hypersensitivity reaction to another beta-lactam agent (e.g., cephalosporin, carbapenem or monobactam).
Drug Interactions
Probenecid
Increases and prolongs amoxicillin blood levels by decreasing renal tubular secretion.
Allopurinol
Concomitant use may increase the incidence of skin rashes.
Methotrexate
Amoxicillin may reduce the renal excretion of methotrexate, increasing its toxicity.
Oral Contraceptives
May decrease the efficacy of oral contraceptives (rarely reported).
Oral Anticoagulants (e.g., Warfarin)
May prolong prothrombin time; INR monitoring is recommended.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal disturbances (nausea, vomiting, diarrhea) and fluid and electrolyte imbalance. In cases of very high doses, crystalluria leading to renal failure has been reported. Management is symptomatic and supportive. Gastric lavage or activated charcoal may be considered soon after ingestion. Hemodialysis can be used to remove amoxicillin from circulation.

Pregnancy: Category B. Amoxicillin has been widely used in pregnancy and is considered generally safe. Use only if clearly needed. Lactation: Amoxicillin is excreted in breast milk in small amounts. Generally considered compatible with breastfeeding, but monitor the infant for diarrhea, candidiasis, or allergic reactions.
