Description
Overview of the medicine
Ancef 125 mg Pediatric Drop contains Cefazolin, a first-generation cephalosporin antibiotic used to treat a wide range of bacterial infections in infants and children. It is effective against both gram-positive and some gram-negative bacteria.
Uses & Indications
Dosage
Adults
Not applicable for this pediatric oral drop formulation.
Elderly
Not applicable for this pediatric oral drop formulation.
Renal_impairment
Dosage adjustment required for significant renal impairment. Consult a physician for specific guidelines based on creatinine clearance.
How to Take
Administer orally using the provided calibrated dropper. Shake the bottle well before each use. Typical pediatric dosage is 25-50 mg/kg/day divided into 3 or 4 equal doses. For severe infections, up to 100 mg/kg/day may be used. Do not exceed 6 g/day.
Mechanism of Action
Cefazolin is a bactericidal antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs) located inside the bacterial cell wall, leading to cell lysis and death.
Pharmacokinetics
Onset
Rapid, within minutes to an hour for antibacterial effect.
Excretion
Primarily renal (glomerular filtration and tubular secretion) in urine.
Half life
Approximately 1.8 hours (adults); can be longer in neonates/infants.
Absorption
Well absorbed orally when administered as a suspension. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Minimally metabolized; primarily excreted unchanged.
Side Effects
Contraindications
- •Hypersensitivity to Cefazolin or any other cephalosporin antibiotics
- •History of severe hypersensitivity to penicillin (due to potential cross-reactivity)
Drug Interactions
Probenecid
May decrease renal tubular secretion of Cefazolin, resulting in increased and prolonged plasma concentrations of Cefazolin.
Aminoglycosides
Concurrent use may increase the risk of nephrotoxicity.
Oral Anticoagulants
May enhance the anticoagulant effect by inhibiting vitamin K synthesis or by displacing warfarin from plasma protein binding sites.
Storage
Store dry powder at room temperature (15-30°C), away from moisture and direct light. After reconstitution, store in the refrigerator (2-8°C) and discard after the specified period (e.g., 10-14 days). Do not freeze.
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and in severe cases, neurological effects such as seizures, especially in patients with renal impairment. Treatment is supportive; hemodialysis may be useful in severe cases.
Pregnancy & Lactation
Pregnancy Category B. Cefazolin crosses the placenta. Use in pregnancy only if clearly needed. Excreted in breast milk in low concentrations; use with caution in nursing mothers.
